Im the one that was paying for Netflix for my family, but the password crack down motivated me to learn how to build a server and go full arrs. They had a good thing going, but now that $26 a month will be used to buy hdds.
I just wanted to see Luke in the role of obi wan or Yoda after decades of Jedi training be a hero and pass on his legacy by training the next generation. But I would have been ok with one heroic lightsaber battle, and a reunion with han, chewy and Leia.
Watching the last Jedi and seeing someone who tossed his blade away because he saw the good in essentially “Space Hitler” try to kill his own nephew because he was having a nightmare so out of character. Then having him overdose on the force and die like a chump, broke me. I left the theater in silence. The last Jedi is also the only star wars movie with out a light saber fight. No blades ever touched.

Pretty good description. Few extra points. -you don’t need to don’t need to do PCR to have enough Nucleic acids to visualize on the gel. Often you use gels to visualize things like purified plasmids. You can also digest your plasmids with enzymes to confirm genomic insert. -these type of gels have many uses outside of direct comparisons, for example you can use the gel purity DNA of a particular size by cutting out the band, melting the gel and using a silica column to pull the DNA out of the melted gel slice. This is super common when you do molecular cloning.
For what is possibly wrong with the top gel.
-the blurred bands can be a lot of things. It might just be inherited to the sample type, for example raw genomic DNA will look like this because you’ll have enzyme that degrade DNA, and a lot of mid replication chromosomes resulting in fragments existing across a spectrum of sizes equally. Additionally the it could also be user error with overloaded well or not enough loading buffer in the sample so it’s not pushed in to gel evenly, or it was run to fast.
Overall the top gel just looks like crap and the bottom gel looks great, but depending on what was run you really don’t know what the gel should look like, if I was testing endonuclease activity the top gel would probably be what I was hoping to get. If was doing PCR on single gene, the bottom one is better.
The reason people are getting stuck up on sports and gendered bathrooms is that it’s a wedge issue that can be weaponized to keep people fighting and distracted by culture war nonsense so the focus isn’t on the building of the Orange Reich, a dozen companies contributing to almost all the climate change, and our economic system working to keep us enslaved.
I paid $1000 for books my first semester of college back in 2007. I felt so burnt and violated I never bought another textbook. I made it through the rest of undergrad, a masters, and a PhD in biochemistry by checking out books from the library, borrowing textbooks from friends, and going sailing. When I taught I made it a point to teach my students about all the ways they can avoid becoming a victim like myself.
The everything is woke or not is out of control, when the real wide spread problem is just bad writing and story telling. I’m just tired of stories with contrived conflicts that occur just to make the story move forward “ie the entire plot of God of war Ragnarok”, and established characters making uncharacteristic choices and actions without any development towards that change, “ie Luke Skywalker in the last Jedi and basically everyone in the last season of game of thrones”.

That’s probably more of a reason to get her vaccinated. Those topical flea and tick meds are rough. We got our cat vaccinated even though she was 100% indoor because she used to get too sick with topicals and liked to cuddle the dog.
Poor thing, I’m glad he has a loving family now. I once found a golden retriever wandering the forest while hiking in Indiana. She had so many ticks on her we had to take her to an emergency vet (since it was Sunday) to get them off. Luckly she was microchipped and we found her family who has been looking for her for over a week at the time.
If only there was a vaccine for Lyme disease so the poor pupper wouldn’t have had to suffer in the first place

I’ve repaired about 20 different Gameboys. The community for Gameboy repair and mods is huge. I suggest watching a few of macho nacho videos on YouTube to work up the nerves to open it. Paying people to review articles is just going to make things worse. The real problem is open access. In the current system the author of the manuscript has to pay to publish it, and the publisher turns around and asks readers to pay to access it. Its a scam. If research was conducted with any public money the knowledge generated should be public. This is why Elsevier needs to go. If you saw how much money institutions have to pump into these useless publishers to get access to knowledge funded by the public for the public there would be more outrage.
